As well as good old fashioned manuscript paper (and Sibelius software), I write music straight onto audio software. Here’s what I tend to use:

Cubase 5 controlling the following sample sets:

East West Symphonic Gold
East West Collossus
East West Stormdrum
East West Quantum Leap RA
Native Instruments FM8
NI Pro-53
NI Battery 3
NI B4 II

And others!

I have a home set-up with midi controllers, and I use a combination of sampled and real instruments to create real-sounding tracks with a quick turnaround. I have a pool of instrumentalists who I can call on to record solo lines to enhance the “real” sound of my output.
